Sažetak
Introduction: The aim of our study was to examine the influence of hyperbaric oxygenation (HBO) on Na+, K+-ATPase activity in optic nerve of ischemic rats. Materials and Methods: Animals: Hannover-Wistar rats, weighing 250 g. Models: Global cerebral ischemia: vertebral arteries were occluded by electrocautery at the level of the first cervical vertebra.The day after, the common carotid arteries were exposed and a forebrain ischemia was produced by tightening the carotid artery clips for 20 min. Na+, K+-ATPase assay: Na+, K+-ATPase activity was determined spectrophotometrically by modification of the method described by Stefanovic et al (1974). Hyperbaric oxygenation: Animals have been exposed to HBO during seven consecutive days, at 222.86 kPa pressure of 99.5% oxygen, lasting 60 min. Procedure: Animals were exposed to global cerebral ischemia and after different reperfusion period (immediately after ischemia, 30 min, 1, 2, 24, 48, 72 and 168 hrs), Na+, K+ -ATPase activity was measured. A part of ischemic animals were exposed to HBO after reperfusion. Results: We found significant decrease in Na+, K+-ATPase activity in periods from 30 min to 7 days after ischemia. The most profound decrease was registered in 24 hrs reperfusion time. Treatment with HBO prevented enzymatic inhibition in animals exposed to hyperbaric oxygen in a period up to 24 hrs after ischemic injury. Conclusions: Decrease in Na+, K+-ATPase activity was evident in ischemic animals exposed to different reperfusion intervals. Treatment with HBO maintained enzymatic activity up to control levels, and exerted neuroprotective effect.
