Sažetak
Relapse after bone marrow transplantation (BMT) remains a significant problem. Previous studies have used standard cytogenetic techniques, changes in red cell phenotype or appearance of both Y chromosomes. In the case when any of these markers is not useful, short tandem repeats (STR) loci can be a potentially tool for the diagnostic. In order to establish the strategy for the follow up of chimerism, in this study 10 patients (before and after BMT) and their HLA identical donors were analysed for six STR loci (HUMTH01, HUMVWA31, HUMFES/FPS, HUMF13A1, SE33, D1S80). DNA was isolated by standard salting out method. After amplification samples were run on 6% polyacrilamid gel and detected automated by a Pharmacia ALFexpres. Assignment of alleles was performed using a Fragment Menager program. In all 10 HLA identical pairs we found differences between recipent and donor for at least two STR loci. Namely, these tested loci have six and more known alleles. A complete chimerism was present in all 10 patients and any of patients was not show relapse of disease. The results of this study showed the application of STR polymorphism for monitoring post-transplant chimerism. Further prospective studies will examine the clinical implications of potential mixed chimerism and the detection of residual haematpoiesis as a sign of early relapse.
