Sažetak
For the first time, Pepino mosaic virus (PepMV) infection of greenhouse tomato, was reported in the Netherlands and UK in 1999. Since then, this viral disease has become a significant problem despite the measures of eradication. PepMV is on the EPPO Alert List. In order to harmonize legislation with the EU, Croatia has established Order on measures to prevent the introduction and the spread of Pepino mosaic virus (O.G. 56/08 and 46/11). An extensive survey among Croatian tomato growers was conducted in 2004. During the last seven years of survey (2004-2010), 1225 samples were tested: 1195 samples from commercial domestic tomato greenhouse production, 28 from imported seedlings and 2 from imported fruits. Samples were analysed by enzyme-linked immunosorbent assay, DAS-ELISA (Loewe) and DASI-ELISA (Neogen). Serologically positive results were confirmed with IC-RT-PCR by the use of PepMV antibodies (Loewe) and two pair of primers (UK/UK1 and TBG/UTR). In the first six years, PepMV was absent in domestic greenhouse tomato production, but in 2010 its presence was confirmed in two counties at three locations (Gajić and Magadenovac in eastern region and Bijela zemlja in western part of country). The origins of the seedlings that became infected were Hungary and Bosna and Hercegovina. The eradication measures were immediately implemented as well as disinfection of space and tools. Also, the growing season was postponed for three months. The surveys will be continued in the future.
