Sažetak
Mannose receptor (MR) belongs to type II C type lectin receptor super family together with DEC205. The MR recognizes surface polysaccharides of several pathogens as well as some endogenous highly glycosilated molecules. It facilitates their binding, uptake and delivering to early endosomes where the antigen is retained without degradation. A little is known about the presence and MR mediated endocytosis at the materno-fetal interface. The aim of our study was to examine presence, distribution and possible endocytic activity of the mannose receptor at the materno-fetal interface. As material we used first trimester normal human decidua following elective termination of 6-10 week old pregnancies. The presence of MR+ cells was analysed by immunohistology of frozen decidual tissue sections by anti-MR IgG1 monoclonal antibody clone PAM-1 (IgG1, PAM-1 MoAb). Immunofluorescency and flow cytometry was used to analyse presence and distribution of MR+ cells in freshly isolated or overnight cultured decidual cell suspensions. Endocytic activity was measured by uptake of Dextran-FITC. We found MR+ cells in first trimester decidual tissue sections with dominate distribution around the glands. Freshly isolated decidual cell suspensions contained about 20% of MR+ cells. The majority (80-90%) of these MR+ cells belong to CD14+ decidual macrophages and about 10% to CD83+ dendritic cells. The total number of MR+ cells significantly decreased following overnight incubation in the medium only. The MR+ cells following overnight culture in the medium only were able to uptake Dextran – FITC. In the first trimester pregnancy decidua CD14+ and CD83+ cell populations express MR and are capable to mediate MR-dependent endocytosis in vitro.
