Sažetak
Due to their high level of polymorphism and reproducibility, microsatellite markers have been widely utilized in genetic diversity and cultivar identification studies. Being species- specific, microsatellite loci have to be identified, isolated, and characterized in the species of interest. The development of microsatellite markers from genomic libraries enriched for specific microsatellites usually involves considerable time and cost. The National Collection of Medicinal and Aromatic Plants currently holds more than a hundred basil (Ocimum basilicum L.) accessions that have been used in numerous studies including genetic analyses based on RAPD and AFLP markers. However, reliable cultivar identification and genetic relationship analysis have been impeded by the lack of microsatellite markers available. By using nextgeneration sequencing technologies, a large amounts of DNA sequence data have been made publicly available, not only for model plants and major crops but also for a wide range of ‘less important’ species including basil. In particular, expressed sequence tags (EST), resulting from whole transcriptome sequencing, provide valuable resources to develop gene- associated microsatellite markers (EST-SSRs). In this way, we plan to indentify microsatellite markers for basil without the need of time consuming and expensive process of genomic library construction.
