Sažetak
Lactobacillus amylovorus strains are attracting interest as potential workhorses to be employed in industrially applicable degradation of raw starch and direct production of enatiomerically pure intermediates thereof, e.g. D- or L-lactate. Besides whole-cell biocatalysis, production of raw starch degrading enzymes is industrially relevant process. The L. amylovorus strains have been used as sources of genes encoding the enzymes since the genes expression, enzyme production and secretion were proven in several species of the same genus. However, available results obtained by transformants and constructs give mild insight into biochemical properties of the enzymes. Wild-type raw starch degrading enzymes were produced by the strain L. amylovorus DSM 20531 in response to maltose as an inducer. The disaccharide was added instead of glucose to standard MRS medium and modified MRS medium was used in the enzymes production at a laboratory-scale. Isolation of the enzymes from broth supernatant and utilization of two starch-type substrates including insoluble substrate was investigated. In both cases the wild-type enzymes contributed to the starch degradation and these reactions yielded mainly oligosaccharides.
