Sažetak
Liver has major role in fat and carbohydrates metabolism and it is one of the organs where the accumulation ectopic fat is taking place. Lipid accumulation in the liver promotes systematic metabolic dysfunction. In this study we analysed lipid accumulation based on functional organisation of liver parenchyma. Study included male and female Sprague-Dawley rats divided in 4 groups: 1. Standard diet (SD), 2. Diet rich in carbohydrates and fat (HFHSD), 3. HFHSD + metformin treatment and 4. HFHSD + liraglutide treatment. The experiment lasted for 20 weeks. Two different methods used to analyse lipid stores in the liver: Sudan black B stain for all lipids and Oil Red O for neutral lipids and fatty acids. There is sex specific pattern of lipid accumulation in the liver, HFHSD significantly increased lipid accumulation only in male rats. The lipid accumulation was less with Liraglutide and metformin. In male rats with HFHSD lipids accumulated with the same intensity in whole liver parenchyma, while in female group accumulation is dominant in peripheral part of the liver. This finding could be related to estrogen protective effect against hepatic fat accumulation by promoting insulin activity on glucose metabolism and limiting hepatic lipid deposition since elderly female rats still produce estrogen. Based on the hepatic acinus functional model the lipid accumulation in the liver is sex specific and zonal (peripheral and central).
