Sažetak
AIM: Previously, we have demonstrated that 3 weeks of intake of omega-3 enriched eggs reduced the level of oxidative stress in young healthy men (1) and had a positive effect on reducing systemic oxidative stress in young healthy women due to increased blood antioxidative capacity (2). Present study aimed to investigate the effects of n-3 polyunsaturated fatty acids (n-3 PUFA) enriched eggs consumption on the levels of oxidative stress in human leukocyte’s subsets. MATERIALS AND METHODS: 33 healthy participants (18 men and 15 women, age: 20-26) included in the protocol were divided into 2 groups: OE group consumed n-3 PUFA enriched chicken eggs (n=18 ; 2 eggs/day, n-3 PUFA=403.10 mg/day) while RE group consumed regular chicken eggs (n=15 ; 2 eggs/day, n-3 PUFA=74.63 mg/day) for 3 weeks. Peripheral leukocytes’ oxidative stress was measured by flow cytometry (BD FACS Canto II flow cytometer ; Flowlogic v7.2.1 software) with DCF- DA (2’, 7’ - dichlorofluorescin diacetate) assay at the beginning and at the end of the study. Levels of intracellular ROS were assessed prior and 30 minutes after PMA (phorbol-12- myristate-13- acetate) (1:1000 ; 50µl) mediated induction of ROS production. Statistical analyses were performed using Sigma Plot v12.0 and GraphPad Prism v6.01 softwares. T-test was used for differences among groups and intragroup differences were tested by paired t-test ; p<0.05 was considered significant. The study protocol and procedures conformed to the standards of the Declaration of Helsinki and were approved by the Ethical Committee of the Faculty of Medicine, University of Osijek. RESULTS: Leukocytes’ baseline and PMA stimulated ROS production did not differ between the study groups at the beginning of the study. At the end of the study, OE group exhibited significantly reduced levels of ROS upon PMA stimulation in male granulocytes (p=0.0442), in contrast to significantly increased ROS production upon PMA stimulation in male and the female monocytes from OE subjects (p=0.0006 and p=0.0227, respectively). At the end of the study, OE men had significantly lower level of oxidative stress in granulocytes prior and upon PMA stimulation (p=0.0366 and p=0.0049, respectively) compared to RE men. At the end of study OE men had significantly lower levels of ROS in monocytes and lymphocytes compared to OE women (p<0.0001 and p=0.0153, respectively). CONCLUSION: Omega-3 consumption reduced levels of oxidative stress upon PMA stimulation in granulocytes of men, while it had an opposite effect on the monocytes of both sexes. Results suggest that the omega-3 supplementation during 3 weeks reduces oxidative stress in granulocytes probably by increasing the cellular antioxidative capacity, and this effect seems to be restricted to male subjects.
