Sažetak
A facultatively fermentative yeast Hansenulta polymorpha CBS 4732 is referred in literature as a yeast that does not ferment disaccharides in oxygen-limited conditions. Results of Durham test confirm literature data showing that H. polymorpha does not ferment sucrose but ferment glucose as control sugar intensively producing carbon dioxide bubbles after three day cultivation. In contrary, growth of H. Polymorpha in chemostat under oxygen limited conditions resulted with significant ethanol production. An active fermentative metabolism on sucrose in such controlled conditions is in contradiction with results of Durham test and literature data. In this work reasons for unreliability of Durham test are discussed and alternative methods for estimation of fermentative ability of yeasts are recommended.
