Sažetak
Mitochondria can exist in two different well-defined physiological states, promitochondrial and functionally fully developed, depending on presence or absence of medium dissolved oxygen. They contain double-membrane system, and therefore require large amounts of membrane components. Aim of this work was to analyze lipid composition of mitochondria of two industrial yeast strains grown under aerobic (baker's yeast) and anaerobic (brewer's yeast) conditions. Baker's yeast mitochondria contained six times more phospholipids (PL) and four times more ergosterol in respect to proteins than brewers yeast did. The main characteristics of phospholipid composition of baker's yeast mitochondria were high content of phosphatidylcholine (44 % total PL), phosphatidylethanolamine (18 % TPL) and relatively large amount of cardiolipin (14 % TPL). In phospholipid composition of brewer's yeast mitochondria phosphatidylcholine was the major phospholipid species, too, but its amount was significantly decreased on account of phosphatidylinositol (30 and 26 % TPL, respectively). Amount of cardiolipin was also significantly lower (10 % TPL). The main characteristics of neutral lipid compositions were high content of ergosterol (39 mg/g proteins) in baker's and high content of squalene (22 mg/g proteins) in brewer's yeast mitochondria. Data prove close relationship between growth conditions, the composition of mitochondrial lipids and physiological state of mitochondria.
