Sažetak
The effect of heating rate, crystallinity (crystalline vs. amorphous) and different particle sizes on thermal behaviour of trehalose as well as of physical mixtures with paracetamol were investigated using various techniques: thermal methods of analysis, hot-stage microscopy, scanning electron microscopy, Fourier-transform infrared spectroscopy and variable temperature X-ray powder diffraction. The study has shown that thermal behaviour of trehalose dihydrate largely depends on particle size, heating rate and pan type used in DSC measurements. Particle size was also found to make a significant effect on process of dehydration of trehalose dihydrate. Results of variable temperature X-ray analysis and modulated temperature DSC have shown that trehalose dihydrate with particle size smaller than 45 mm undergoes glass transition after the dehydration, in contrast to particle size larger than 500 mm. Particle size as well as heating rate also makes a great influence on thermal behaviour of physical mixtures of trehalose with paracetamol. Presence of paracetamol in physical mixture was found to enhance crystallisation of anhydrous trehalose by depressing temperature of anhydrous trehalose formation.
