Sažetak
There is increasing number of studies using inflammatory mediators as biomarkers for skin inflammatory diseases. In our previous studies Luminex multi-assay technique was used to explore candidate biomarkers for UV-induced inflammation in the skin and in atopic dermatitis. Using multi-array assay based on electrochemiluminescency (Mesoscale Discoveries) several candidate biomarkers of local immune response in the skin have been identified and the method showed to be more sensitive than Luminex or ELISA assay. For the purpose of analytical evaluation of Mesoscale technique, additional skin samples of patients of different phenotype and filaggrin genotype and healthy individuals were sampled. The method was evaluated in regard to extraction procedures and its dynamic range, sensitivity, limit of detection and quantification, intra- and inter-assay variability under different experimental conditions.
