Fatty acids are essential components of membrane phospholipids having therefore an important role in maintaining of a structure and a function of cell membranes. Accumulation of free arachidonic acid, C20:4, precursor of prostaglandins and leukotriens, is an indicator of nervous cell damage caused by neurotrauma. In this study we have followed the effect of indomethacin, a potent inhibitor of endogenous prostaglandin synthesis, on the level of arachidonic acid in an experimental model of spinal cord injury in rabbits. Indomethacin was tested in the doses of 0.1 ; 0.3 ; 1.0 and 3.0 mg/kg of body weight. Spinal cord samples from the impact injury site were taken and frozen in liquid nitrogen. Modified method by Folch, for the isolation and purification of total lipids, was used. The FFAs were separated from the total lipid extract by preparative thin-layer chromatography. Arachidonic acid was identified and quantified using gas chromatography. The level of arachidonic acid was increased in the spinal cord of experimental animals after neurotrauma. Indomethacin treatment induced significant decrease of the free arachidonic level caused by neurotrauma.
