Sažetak
Introduction: Extracellular Hsp70 (eHsp70) was shown to be increased in sera of patients with lung diseases, such as chronic obstructive pulmonary disease (COPD). Some studies demonstrated that eHsp70 can act pro-inflammatory and trigger sterile inflammation via Toll like receptors (TLR) 2 and 4. Aims and objectives: We hypothesized that eHsp70 might induce inflammatory effects by provoking cytokine secretion. Therefore, we assess concentrations of interleukin (IL) 6, IL-8, and tumor necrosis factor α (TNF-α) in NCI-H292 cell line, used as a model of tracheo-bronchial airways in COPD. Methods: NCI-H292 cells were treated with 0.1, 0.3, 1, 3, 10 and 30 µg/mL low endotoxin human Hsp70 recombinant protein (rhHsp70) for 24 h. Cytokines’ concentration was measured in cell supernatants by ELISA method. Results: Significantly increased secretion of both IL-6 and IL-8 was found with 0.3, 1 and 3 µg/ml rhHsp70, while IL-6 was also increased with 0.1 and 30 µg/ml rhHsp70. Secretion of TNF-α was not induced with rhHsp70. Conclusion: The obtained results show that eHsp70 has strong pro-inflammatory effects on NCI-H292 cells and therefore might trigger an inflammatory process in COPD.
