Sažetak
Problem Decidual NK cells (dNK) of CD3-CD16-CD56bright+ phenotype are dominant cell population infiltrating first trimester decidua. Decidual NK are rich with cytolytic mediator-perforin. The aim of this study was to analyse the effect and possible mechanism of progesterone's regulation of cytolytic potential and activity of DL against NK sensitive K-562 targets. Methods of Study Decidual cells were isolated by enzymatic digestion of first trimester human decidua and stimulated for 18 hr in vitro. DL were isolated as nonadherent cell fraction and their cytolytic activity against K-562 targets were determined by PKH-26 (red) 2h cytotoxicity assay and analysed by flow cytometry. Immunofluorescency, immunohistology and immunocytochemistry were used to analyse perforin+, PIBF+ and IL-15+ cells. Results DL in vitro lyse about 40% of K-562 targets by perforin mechanism. Progesterone and its mediator Progesterone Induced Blocking Factor (PIBF) decrease cytolytic activity by making perforin stay in the cells. Freshly isolated CD56+ dNK are PIBF+. Interleukin-15 upregulates perforin expression and cytolytic activity of DL against K-562 targets and it is found in first trimester pregnancy decidua. Progesterone and PIBF decrease percentage of IL-15+/HLA-DR+ decidual cells. IL-15 reverted progesterone/PIBF mediated suppression of cytolytic activity of DL against K-562 targets. Conclusion It is possible that mechanism of progesterone/PIBF mediated suppression of cytolytic activity of DL against K-562 cells includes decrease of IL-15 cytokine production in decidual macrophages.
