Sažetak
Perforin (P) is a cytolytic molecule located in granules of cells mediating contact-dependent cytotoxicity. The number of P+ cells in decidua of pregnancy is larger than observed in any other, even pathological condition. P-expressing decidual lymphocytes (DL) are prevalently nonclassical NK cells (CD56bright+ CD16-) with low cytolytic activity but fully equipped with potent cytolytic machinery (Pbright+). In the decidua they are distributed randomly as are decual macrophages. P expression upon in vitro cultivation and stimulation was studied at the protein level in peripheral blood lymphocytes (PBL) of women in the first trimester of pregnancy, in decidual lymphocytes, and their subpopulations. Both the percentage of P+ cells and average fluorescence intensity (AFI), as a measure of P content/cell, decreased profoundly during 18 hours of incubation. These changes could not be prevented by stimulating PBL with anti-CD3 mAb, PHA and/or IL-2. However, the disappearance of P+ cells and decreased of AFI for P could be prevented completely by cocultivation of PBL with syngeneic or allogeneic decidual macrophages. The same phenomenon was observed for DL. The analysis of subpopulations showed significant increase of CD56+ cells and total P+ cells, as well as CD8+P+, CD56+P+ and CD25+P+. It is interesting that disappearance of P in the culture of DL was paralleled by increase in percentage of cells expressing Fas ligand. However, Fas ligand expression was prevented when DL were cultivated with decidual mecrophages.
