Sažetak
Background: Isoflurane postconditioning cause an early increase in progenitor cells number, however, in the chronic phase of infarct healing, this number was found to be smaller, suggesting positive effect on infarct scar maturity. The myofibroblasts role is crucial in an early phase infarct area contraction, however, their number in mature scar is minimal. The main objectives were to see whether isoflurane post-conditioning stimulate progenitor cells differentiation to myofibroblasts and to verify hypothesis of more mature looking chronic scar area. Methods: Ischemia was induced for 30 minutes on Sprague- Dawley female rats. Starting from the last 5 minutes of ischemia up until 10 minutes into reperfusion time, the isoflurane group (n=8) received 1.5% of isoflurane, while the control group (n=8) received only an air/oxygen mixture. Immunohistochemistry was performed for infarct area analysis. Results: In subacute phase of infarct healing the number of myofibroblasts (2431.6 ± 331.3) was higher in isoflurane-treated animals compared to the control group (1321.8 ± 293.5 p=0.002). In chronic phase of infarct healing postconditioned animals had smaller number of myofibroblasts (197.5 ± 47.1 vs. 290.0 ± 57.1, p=0.046), even those ones derived from progenitor cells (23.3 ± 4.6 vs. 40.7 ± 8.6, p=0.011), along with higher percentage of mature blood vessels (61.2 ± 2.9 vs.45.5 ± 4.3, p=0.029). Conclusions: Isoflurane-treated animals had higher number of myofibroblasts in granulation tissue. The smaller quantity of myofibroblasts along with the higher number of mature blood vessels found in chronic phase of healing indicate the faster healing of infarct area of isoflurane-treated animals.
