Sažetak
The genomes of Pag sheep and donkey subpopulations in Croatia. Were typed using several methodological approaches. In donkeys we estimated frequencies of transferrins and albumins in addition to typing of eight microsatellite loci. Genetic distances of donkey subpopulations based an to frequencies of polymorphic proteins match were the distances based on microsatellite typing. We sequenced 417 bp long fragment of the proksimal part of the D-loop region of donkey mtDNA. We found two basic sequence types within which several subtypes are present. The phylogenetic separation of two main sequences (X and Y) happened in a period 1, 2 to 3, 3 million years ago. We analysed the same segment of the D-loop region by PCR-SSCP and detected in addition to two main variants also an additonal subtype X'. In the Pag sheep breed we determined frequencies of hemoglobine, transferrin and albumin. We typed seven microsatellite loci and determined a high degree of variability. The average number of allele variants per locus was 8, 71. We sequenced 400 bp long fragment of the D-loop region. The existence of two basic types of mtDNA sequences was confirmed. The rarer sequence type (M) is genetically close to the sequence of Merino breed deposited in GenBank (AF010406) which draws to the conclusion that it might he a trace of the Merinisation of the Pag sheep. The other sequence type (N) belongs to the autochthonous Pag sheep. Frequencies of A and B variants of β -lactoglobulin (0, 5375 ; 0, 4625) and A and D variants of α s1 casein (0, 0875 ; 0, 1250) showed that by selection pressure on certain genotypes the efficiency of cheese production could be improved. These results are the first indicators for genetic profile of mentioned breeds and will stimulate further research and application of molecular data for selection.
