Sažetak
In this study we performed analysis of HLA polymorphism with the goal to identify the presence of rare alleles among donors asigned to the Croatian Bone Marrow Donor Registry (CBMDR). HLA-typing for HLA-A, -B, -DRB1 loci was performed using the PCR-SSO method with a Luminex system and Gene-Probe kits. Less frequent alleles were confirmed at high resolution level using adequate Olerup SSP comercial kits. In the cohort of 4000 individuals, 28 alleles (10 at HLA-A, 11 at HLA-B and 7 at HLA-DRB1 locus) were observed at frequency of 0.01%: A*01:02, A*01:03, A*02:11, A*02:12, A*02:45, A*03:16, A*25:02, A*36:02, A*66:02, A*74:01, B*13:10, B*14:03, B*27:09, B*27:12, B*27:30, B*39:10, B*39:15, B*44:29, B*50:02, B*51:29, B*57:08, DRB1*04:10, DRB1*04:13, DRB1*11:06, DRB1*13:15, DRB1*14:05, DRB1*15:07 and DRB1*16:05. Among remaining 28, 000 volunteer donors in CBMDR, additional 41 HLA alleles with frequency <0.01% were observed: 8 alleles at HLA-A locus (A*02:17, A*02:30, A*02:60, A*03:108, A*24:17, A*31:08, A*68:06, A*68:09) ; 17 alleles for HLA-B locus (B*07:10, B*07:02:28, B*08:35, B*15:04, B*15:07, B*15:23, B*15:39, B*14:11, B*37:09, B*39:40N, B*40:08, B*40:09, B*44:04, B*44:06, B*51:09, B*53:05, B*78:08) ; and 16 alleles at HLA-DRB1 locus (DRB1*03:02, DRB1*03:04, DRB1*03:06, DRB1*04:16, DRB1*08:06, DRB1*08:18, DRB1*11:13, DRB1*11:15, DRB1*11:16, DRB1*11:28, DRB1*11:56, DRB1*11:58, DRB1*13:21, DRB1*13:23, DRB1*13:32, DRB1*14:02). According to the Allele Frequency Net Database (www.allelefrequencies.net), alleles A*68:06, B*14:11, and B*37:09 are defined as rare, while alleles B*07:02:28 and B*39:40:01N are defined as very rare. The data provided by our study give insight into the genetic diversity of the HLA system in the Croatian population and should be of benefit in future donor searches as part of the unrelated HSCT program.
