Sažetak
Registries of volunteer hematopoietic stem cell donors and umbilical cord blood (UCB) banks contain valuable data for studying allele and haplotype frequencies in the general population. HLA-typing for A, B, DRB1 loci of the 644 UCB units banked in Croatian Cord Blood Bank (CCBB) and of the 12173 blood samples from Croatian Bone Marrow Donor Registry (CBMDR) was performed at medium resolution level using the SSO method with a Luminex system and One Lambda kits. Typing of UCB units revealed 18 allelic variants for A locus, with A*02 (27.01%), A*01 (11.80%) and A*03 (10.71%) as the most frequent ones. The number of variants for B locus was found to be 28 and the most frequent were B*35 (13.04%), B*44 (10.3%) and B*51 (9.9%). Locus DRB1 showed the least polymorphism with 13 allelic variants observed among which the most widespread were DRB1*11 (15.5%), DRB1*01 (10.9%) and DRB1*13 (10.3%). A total of 11 A, B, DRB1 alleles (4, 6, 1 respectively) have been found at the frequency <0.5% and consequently considered as rare alleles. All data on the allele frequencies from UCB correspond well with the frequency data from CBMDR. We have also analyzed haplotype frequencies in both studied groups. The most frequent haplotypes were A*01-B*08-DRB1*03, A*03-B*07-DRB1*15, A*24-B*44-DRB1*07. Furthermore, analysis of genotypes incorporating one or more rare alleles enabled us to identify several rare haplotypes: A*80-B*45-DRB1*09 ; A*80-B*58-DRB1*03 ; A*02-B*73-DRB1*04 ; A*69-B*15-DRB1*03 ; A*34-B*14-DRB1*04 ; A*24-B*54-DRB1*14 ; A*26-B*48-DRB1*12. The characteristics of HLA-A-B-DRB1 allelic and haplotypic polymorphism in Croatia will be helpful in finding matched donors for patients and provide foundation for further studies in population genetics.
