Sažetak
Flavonoids are present in natural products such as fruits and vegetables and have numerous health benefits particularly with respect to their reported antioxidant properties [1]. Quercetin (3', 4', 5, 7-Tetrahydroxyflavan-3-ol) is a member of flavonoids and due to its positive impact on human health it can be used as nutraceutical or food supplement [2]. Such flavonoid compound can be extracted from natural products using a variety of solvents, among them water [3]. The solubility of a substance fundamentally depends on the physical and chemical properties of the solute and solvent as well as on temperature, pressure and the pH of the solution. In this study we demonstrated the water solubility of quercetin at different temperatures (25-100 °C) and time (2-60 min). The thermal degradation of quercetin was monitored on LCMS/MS. The aqueous solubility of quercetin varied from 0.409 ppm at 25 °C to 16.139 ppm at 100 °C. Different heating time did not affect the solubility of quercetin at 25 °C. At 80 °C quercetin solubility was increased with time, 8.248, 10.866 and 11.063 ppm at 2, 30 and 60 min, respectively. However, at 100 °C increasing the heating time resulted with lower amount of quercetin since the degradation of quercetin was more pronounced at that temperature. Protocatechuic acid, 2, 4, 6 - trihydroxybenzoic acid and 3, 5 - dihydroxybenzoic acid were detected as degradation products at 50, 80 and 100 °C and it was showed that increasing temperature and heating time yielded higher amount of detected degradation products. The presented data could be helpful for designing and optimization of extraction of quercetin from natural products using water as ‘‘green” extraction solvent thereby replacing toxic organic solvents.
