Sažetak
Human serum albumin (HSA) is the most abundant protein in human serum. It has numerous functions, one of which is transport of small hydrophobic molecules, such as metabolites, hormones, and nutrients, as well as drugs and toxins. This promiscuous, nonspecific affinity can lead to competition and displacement of different ligand in binding to HSA and cause increase in toxicity of displaced ligands. In this paper, the most important HSA binding sites are described and the risk of displacement interactions between different ligands is discused. While such interactions are more common between physiological (e.g. fatty acids, bilirubin, heme etc) and patophysiological ligands (e.g. indoxyl sulfate, indol-3-acetate, hypuric acid, and CMPF) and drugs, displacement interactions between drugs are also known. In certain cases these interactions can directly influence drug’s free, active concentration in blood.
