Background and purpose: The possible relationship between the level of proinflammatory agent nitric oxide (NO) in human saliva and the presence of the periodontal disease was examined, Also, we examined the influence of conversion of nitrate from food on concentration of nitrite in saliva. Materials and Methods: The production of NO was determined by measuring the concentration of its stable product - nitrite (NOz-) in saliva in 24 patients with chronic periodontitis and in 17 healthy subjects. The nitrite concentration was determined colorimetrical1y by using Griess reaction. The level of NOz- was also determined in both groups of examiners after they have ingested nitrate-rich vegetable juice (red-beet). Results: Mean NO concentration was lower in patients with periodontitis than in control group (p < 0, 01). The concentration of NOz- in healthy subjects, determined for three consecutive days, did not vary significantly. On the other hand, the content of NOz- determined 1.0 to 1.5 hours after ingestion of nitrate rich vegetable juice increased several times in both groups. Conclusion: The reasons for decreased concentration of NO in saliva of patients with periodontal disease are not understood. Conversion of nitrate from food in nitrates in saliva is probably slight in standard diet, but can be significant after ingestion of food rich in nitrates.
