Sažetak
Cells that dye by physiological mechanisms often undergo characteristic changes termed apoptosis. Unlike necrosis which appears to be a passive death, the process of apoptosis is usually considered an active death that requires participation of cellular processes. Given the power of molecular biological and genetic techniques, it is possible to identify genes whose expression changes with a cell's decision to commit suicide, although some examples of apoptosis, that occur in the absence of new gene expression were found. Two classes of genes involved in the process of apoptosis can be distinguished, cell death genes and cell death-associated genes. The identification of cell death genes that are directly involved in cell death regulation, has relied on genetic manipulations that disrupt gene expression. In most cases, blocking the expression of such essential genes saves the life of otherwise condemned cell. For example, tumor suppressor gene p53 and the photo-oncogene c-myc, null mutations in mice, blocking some forms of T-cell death. In contrast, bcl-2 gene offer protection from naturally occurring cell death.Death-associated genes are those whose expression is dramatically up-regulated in dying cells but that are not in themselves lethal.Therefore, they presumably act to facilitate the demise of a cell.
