Different tissues of cactus Mammillaria gracillis Pfeiff. were analysed in in vitro culture. The cactus plants were propagated in vitro on MS medium without any growth regulators. At the bases of some plants as well as of hyperhydric, malformed shoots spontaneously occurred. The aim of this work was to obtain information about causes and conditions leading to spontaneous callus growth in the tissue culture of Mammillaria gracillis Pfeiff. and to investigate regeneration potential of the habituated callus tissue in order to contribute to better understanding of cell differentiation and dedifferentiation mechanisms. Lower agar and higher MS salt concentration. In order to compare habituated callus with tumour, two strains of Agrobacterium tumefaciens were used for transformation of cactus tissue. Two tumours (TW and TR) are hormone-independent and have no organogenic potential. Tumours grew more intensively than the habituated callus. The content of the pahotosynthetic pigments was reduced in hyperhydric tissues of callus and regenerants being even more reduced in TW and TR tumours. The highest peroxidase and esterase activity was determined in transformed tissues. Soluble cell proteins were separated by means of SDS-PAGE and 2-D electrophoresis. A few tissue-specific bands (16, 18, 48 do 50, 56 do 60, 54, 70 and 82 kDa) were noticed. Two-dimensional electrophoresis showed more tissue-specific proteins and differences in glycosylation among different tissues. The glycoprotein of 42 kDa detected by Con A was characteristic only for undifferentiated tissues (callus, TW and TR tumours) and hyperhydric regenerant. Sialinic acid was found in glican component of 42 kDa protein isolated from TW tumour.
