Sažetak
OtcC encodes a hydroxylase that acts at the 6-position of the tetracyclic nucleus, after it has beenformed. OtcC is located at the edge of the otc cluster, distant (and transcribed separately) from the genes for biosynthesis of the polyketide backbone. Disruption of otcC resulted, surprisingly, in a series of incorrectly-folded shorter polyketides (we had expected the tetracyclic structure without a 6-hydroxyl, which has been characterized previously in mutants studied in the 1960's). When a catalytically-inactive mutant version of otcC was used to replace this disrupted copy, the ability to make tetracyclic structures was restored. We hypothesize that OtcC is required as a structural component at the quaternary level to ensure fidelity of the polyketide synthase complex in generating the tetracyclic nucleus.
