Sažetak
Polynuclear complexes of ferric iron with polymeric carbohydrates like dextran or sugars like sucrose or gluconate, used for the treatment of anemia in various diseases, represent a possible source of nonferritin-bound, redox active iron in blood plasma. This iron, “loosely bound” to surface of proteins, DNA, and other macromolecules or chelates, is implicated in oxidative stress and cell injury. It can undergo metal mediated Haber-Weiss reaction or may be reduced to Fe(II) state by ascorbic acid and enter a Fenton-type reaction, in both cases establishing a redox cycle that continuously produces OH• radicals. OH• being a powerful oxidizing agent that reacts at a high rate with most organic and inorganic molecules, scavenges the plasma low-molecular-weight antioxidant (LMWA) pool – its first line of defence against oxidative damage. Depletion of plasma antioxidants due to administration of parenteral iron as an erythropoiesis-stimulating agent in the case of chronic renal failure patients, has been evaluated previously using a variety of techniques including biochemical, immunohistological and spectroscopical. In the present study, the electrochemical method of cyclic voltammetry (CV), that provides information on the LMWA capacity, has been used to detect the change in the oxidative status of iron deficiency anemia patients introduced by i.v. infusion of ferric gluconate. For comparison, spectrophotometric ferric reducing antioxidant power (FRAP) method has also been used. Both, CV and FRAP showed a decrease (p< 0.05) in plasma antioxidant activity between samples collected immediately before and 10 minutes after i.v. infusion of ferric gluconate.
