Sažetak
Prunus spinosa L. (blackthorn), a spiny shrub native to Europe, western Asia and northwest Africa, is used in traditional medicine as a remedy for various diseases. To validate claimed anticancer properties, Prunus spinosa L. blossom alcoholic extract was subjected to bioassays in mouse (Hepa 1-6) and human (Hep G2) liver cancer cell lines. Cells were exposed to different concetrations of extract (10 - 200 μg mL-1), and antiproliferative activity was determined by Kenacid blue method after 72 hours. Inhibitory effects on Hepa 1-6 cells were observed at all concentrations of blossom extract and statistically significant inhibition of cellular proliferation was detected at ≥ 100 μg mL-1 (p<0.025 – p<0.01). Dose dependent significant cytotoxicity of Prunus spinosa L. was also confirmed in Hep G2 cells with concentrations above 50 μg mL-1 (p<0.01 – p<0.001). The extent of the cellular viability loss and accelerated cell death were greater in human vs. mouse liver cancer cells as confirmed with determined IC50 values: 68.2 μg mL-1 in Hep G2 cells < 85.41 μg mL-1 in Hepa 1-6 cells. Furthermore, to assess antioxidant/prooxidant effects of blackthorn blossom extract, ROS (Reactive Oxygen Species) formation was evaluated and results confirmed prooxidant behaviour of Prunus spinosa L. blossom alcoholic extract in both liver carcinoma cell lines.
