Sažetak
Turopolje pig is an autochthonous croatian breed. Historical evidence suggest that it originated through wild boar domestification and partly from other european breeds. Presently, there are only around 250 surviving Turopolje pigs, and this breed is, according to FAO, critically endangered. As a part of an effort to preserve domestic animat biodiversity, a conservation project for this breed is being carried out in Croatia. Microsatellite genotyping has in the past 10 years become the core method for genetic biodiversity study. Microsatellites are ubiquitous and randomly distributed in the genome, they show substantial variation in the number of repeats, and are relatively easily amplified by PCR. The repeat length variants are inhereted as alleles. Length of the alleles can be deterlnined simply and accurately by using automated DNA sequencers. The aim of our research is to establish a genotype profile of the Turopolje pig breed and to determine both within - breed genetic variability and genetic distances from other breeds. For this purpose, we decided to use 12 microsatellite loci tkat esere chosen from the list of 50 microsatellite markers recommended by FAO for genetic diversity studies in pigs. So far genomic DNA has been isolated from 0, 5 ml of full blond taken from 35 Turopolje pigs and 10 hybrid pigs. Microsatellite loci CGA, 50355 and 50090 esere amplified from pigs' DNA by PCR using specific primers. One of the primers in the pair was flourescently labeled by Cy5 on the 5' - end. Resulting DNA fragments esere separated and scored on the ALFexpress DNA sequencer. Data esere analysed by Fragment Manager software. The work is still in progress and the preliminary results already show low heterozigosity for the three analysed loci in the Turopolje pig breed. This is consistent with our expectation because this breed recently experienced a severe demographic bottleneck event.
