Sažetak
Memantine hydrochloride is pharmaceutically active substance that was found to have role in treatment of Alzheimer's disease. Because needle- shaped crystals of mementine hydrochloride were not appropriate for further use in this particular case it was necessary to find another salt of more suitable formation. After reviewing previous research, memantine hydrogen sulphate was chosen since it has significantly better crystal morphology and hygroscopic behaviour. In this research, through the batch crystallization process, the influence of process parameters and addition of auxiliary components (antisolvent and seed) on granulometric properties of memantine hydrogen sulphate was investigated. Type of solvent and process control influence the saturation of the system, thermodynamics, crystallization kinetics and granulometric properties of crystals. Since the aim of this study was to create a product of appropriate granulometric properties, batch crystallization of memantine hydrogen sulphate by cooling from 2- propranol with addition of antisolvent (ethyl acethate and n- Heptane) and seed within metastable zone was performed. In addition, the filtration test was performed and drying kinetics was determined from data obtained by drying in atmospheric dryer. The shape of crystals was observed with microscopy analysis and the distribution of crystal size was determinated using laser diffraction method. Crystallization kinetics was described using appropriate potential equations.
