Sažetak
Mitophagy, a selective autophagy process whereby dysfunctional or impaired mitochondria are cleared from the cell, is very important since inability to remove damaged mitochondria could result in their accumulation and subsequently lead to the development of serious diseases such as cancer or neurodegeneration. Our discoveries of mitophagy receptors, Nix and Bnip3, have shown that they interact with Atg8 proteins in LIR-dependent manner and recruit autophagic machinery to damaged mitochondria. Moreover, we have shown that Nix mediates mitochondrial clearance during reticulocyte differentiation. What are the mechanisms that activate mitophagy receptors is still not fully understood but we postulate that, as other autophagy receptors, these are activated by phosphorylation of the serines adjacent to LIR motif. Further, we propose that Nix and Bnip3 could increase activation of mitophagy by their oligomerization. In our current research, we are investigating how oligomerization status of the receptors influences mitophagy initiation and progression and what is the interplay between phosphorylation and oligomerization status of the receptors.
