Sažetak
Francisella tularensis is a facultative intracellular Gram-negative bacterium that causes tularaemia, a fatal disease, in animals and humans. This organism has been found in various wildlife species, like birds, fish, amphibians, voles, ticks and amoeba. It resists harsh environments, and has been shown to survive in water and mud for more than a year. There are four known subspecies of F. tularensis: tularensis (type A), holarctica (type B), mediasiatica and novicida. The aim of the study was to follow survival of F. tularensis subsp. novicida after exposure to air drying over 60 seconds, 30, 60 and 90 minutes. Drying experiment revealed that the bacterium viability was protected only at 60 seconds of drying. Based on these results we decided to recover the bacterium that was exposed to drying at 60 seconds and 30 minutes by infecting D. discoideum cells. The recovery was followed at 2, 24, 48 and 72 hours by colony forming units on buffered-charcoal yeast extract agar. Results from this study show that external factors, such as drying, damage the viability of cells. The bacterium recovered in amoeba cells only after exposure to 60 seconds of drying.
