Sažetak
Gliomas are tumors originatting from astrocytes, oligodendrocytes or ependimal cells. Those of astrocytic origin are the most widespread of primary brain tumors and account for more then 40% of all CNS neoplasms. The current state of knowledge on the associations between tumor aetiology and oxidative stress allows to conclude that enviromental factors which cause oxidative stress could both induce and promote cancer, especially in case of hereditary predispositions. The aim of present study is to investigate the immunohistochemical expression of 4 HNE-modifying proteins as results of lipid peroxidation in different type of astrocytoma. Our studies were based on 45 astrocytic tumors. These tumors were graded in accordance with the WHO classification and ran to 15 DA, 15 AA and 15 GB. The tumors came from tumor bank material in the form of formalin- fixed, paraffin-embedded tissue. Slides were stained with hematoxylin-eosin and two were prepared for immunohistochemistry with monoclonal antibodies to HNE-hystidin conjugate. Immunohistochemical positivity reaction to HNE was analysed semiquanitativly. HNE positivity was proportional with malignancy of astrocytomas. The lover was in DA, followed by AA and GB. Lowest intensity of HNE immunopositivity was present in tumor cells of almost all DA, predominantly around blood vessels. In more malignant variant of astrocytoma, AA and GB, HNE positivity was moderated to strong, and diffusely distributed in all tumors.
