Cyanobacteria cultivation has found its application in many areas, such as biomass production, production of industrial polymers, metabolites, in dedesertification and agricultural applications, etc. We have investigated the impact in variation of different factors on the development of Nostoc sp. under the laboratory conditions. Nostoc culture was isolated from the environment (luvisol) and cultured on the surface of the soil in petri plates. Two soil types were used in the experiment (luvisol, gley), three light levels, three inoculum concentrations and two sonication levels. Gley supported the development of three times higher chlorophyll concentration compared to luvisol, while total biomass increase was similar between soil types. Total biomass was proportional to the inoculum concentrations and to some degree to light levels on both soil types. Interestingly, chlorophyll concentration on luvisol was highest at the medium light level. The interaction of the light level and the soil type was confirmed by the permutational multivariate analysis of variance. The effects of sonication were not conclusive. The selection of the soil type for the cultivation of Nostoc was the most important factor, followed by the inoculum concentration. Light intensity seemed to be a less important factor. These results improve further investigations of the soil cyanobacteria under the laboratory conditions.
