Sažetak
Some classes of anticancer antibiotics exhibit their antiproliferative activity due to metabolic formation of quinone methides (QMs) that can alkylate biologically important molecules (DNA and enzymes) [1]. However, to date, anticancer activity of simple quinone methides has not been thoroughly investigated, mostly because of their short lifetimes. A photochemical dehydration reaction of suitably substituted phenols provides a very mild and easy approach to QMs in biologically systems [2]. Herein we report synthesis of several bisQMs precursors that are anticipated to induce DNA cross-linking on photoexcitation. Precursors 1 - 10 were prepared by a Grignard reaction of appropriate dibromoalkane and protected benzene or naphthalene o- hydroxycarbaldehyde, which had to be synthesized. The target final bisphenol and bisnaphthol derivatives are new molecules that up to now have not been described in literature so they were characterized by spectroscopic methods. In addition, in vitro antiproliferative activity of the prepared bisQM precursors is going to be investigated.
