Sažetak
A novel cascade reaction for the production of aldol adduct (3S, 4R)-6- [(benzyloxycarbonyl)amino]-5, 6-dideoxyhex-2- ulose, a precursor of D-fagomine, was studied in this work. D-Fagomine was found to have inhibitory activity against mammalian intestinal glucosidase and galactosidase and is useful in reducing the risks of developing insulin resistance and of becoming overweight. The strategy devised consisted of the simultaneous use of three enzymes: horse liver alcohol dehydrogenase (ADH) used for the oxidation of N-Cbz-3-aminopropanol to N-Cbz-3- aminopropanal, D-fructose-6-phosphate aldolase (FSA) variant A129S for the aldol addition of dihydroxyacetone to N-Cbz-3-aminopropanal, and a third enzyme, NADH oxidase (NOX) for coenzyme NAD+ regeneration. This cascade presents an interesting way to obtain aldol adduct using stable starting and readily available materials such as aminoalcohols. The reaction resulted in 76% yield on aldol adduct.
