Sažetak
Coscinasterias tenuispina is a fissiparous starfish, capable of both sexual and asexual reproduction. When reproducing asexually, it splits in half, thus creating two separate individuals. Each of these will regenerate the missing half of the body, in a complex process which is not fully elucidated, resulting in two new individuals capable of further reproduction. The subject of this thesis is to explore the aging and regeneration pattern in these clonal animals, which represent an interesting model for aging studies. Also, it has been suggested that some populations of C. tenuispina can proliferate without sexual reproduction, through fission only. If this is the case, then either these populations do not age, or they are capable of reversing the damage accumulated with age, meaning that they are capable not only to regenerate but also to rejuvenate. To test these possibilities, several well-known biomarkers of aging such as activities of catalase, superoxide dismutase, glutathione and proteasome were measured in both regenerating and non- regenerating arms. The results of measurement of all four parameters show a tendency of their decreasing with length of arm. The actual age of these individuals is unknown, so length of arm is used as a parameter that most likely correlates with their chronological age. Therefore, these results suggest that these starfish do indeed accumulate age-related damage. In highly asymmetrical individuals, there is a significant difference in measured parameters between regenerating and non-regenerating arms, implying that regenerating arms are younger than their non-regenerating counterparts. This might indicate rejuvenation on the level of tissue. In this experiment it was not possible to demonstrate that rejuvenation additionally occurs on cellular level. However, the possibility that some starfish populations can survive only through asexual reproduction implies the capability of cellular rejuvenation, and this possibility should be studied further.
