Sažetak
Activities related to petroleum industry often lead to hydrocarbon contamination of soil and water. For many years, mechanical and chemical methods have been the only way for removing hydrocarbons from contaminated sites. Since these methods often have limited effectiveness and can be very expensive, the aim is to replace them with more efficient and cost-effective methods, such as microbial degradation by indigenous microorganisms in water and soil capable of degrading hydrocarbon contaminants. In this work, metagenome from oil-polluted sea sediments was analyzed using bioinformatic methods with the aim of assessing the biopotential of microbial communities for remediation of hydrocarbon contamination. Targets were enzymes involved in aerobic degradation of short- and long-chain alkanes as well as enzymes for anaerobic alkane degradation. Analyzed metagenome contained enzymes both for aerobic and anaerobic alkane degradation, with majority of genes of interest belonging to alkB alkane hydroxylase and cytochrome P450 alkane hydroxylase families. Selected complete genes from every family of interest will be sent for synthesis, expressed and characterized with the final goal of assessing their potential for bioremediation of oil-polluted sea and soil.
