Sažetak
Aminoacyl-tRNA synthetases are responsible for the specific aminoacylation of transfer RNAs. According to their structural features, the members of this enzyme family can be partitioned into two classes. The active site domain of all so far crystallized class II aminoacyl-tRNA synthetases exhibit an original fold, constituted by several antiparallel beta- sheets, connected by the loops of variable lenghts. In order to study structure/function relationship in yeast seryl-tRNA synthetase (SerRS), which has not been crystallized yet, a number of mutants have been generated by extensive in vitro mutagenesis of SES1 gene and subjected to functional in vivo and in vitro analysis.
