Sažetak
Ochratoxin A (OTA) and citrinin (CTN) are nephrotoxic mycotoxins frequently found together in food and feed. While OTA is one of the most studied mycotoxins, much less is known about the mechanism of CTN toxicity. The purpose of our study was to check the accumulation of OTA and CTN in target organs when given together. Adult male Wistar rats (N = 6) were treated either with OTA (0.125 and 0.250 mg kg−1 b.w., p.o., 21 d) or CTN (20 mg kg −1 b.w., p.o. 2 d) given separately or administered together. The concentrations of OTA and CTN were measured using LC- MS/MS. The increase of OTA concentration in kidney (1.55 ± 0.03, 3.03 ± 0.06 g g−1 tissue) and liver (1.05 ± 0.42, 3.96 ± 0.27 g g−1 tissue) correlated with OTA doses (0.125 and 0.250 mg kg−1 b.w.). When the animals were treated with OTA+CTN (0.125 + 20 and 0.250 + 20 mg kg −1), the kidney concentrations of OTA were lower (1.00 ± 0.26, 0.80 ± 0.12, respectively) than in animals treated with OTA only. The same was true for liver OTA concentrations (0.94 ± 0.40, 1.56 ± 0.74) in OTA+CTN treated animals. Resveratrol (RSV) did not have a protective role in OTA accumulation either in the kidney or in the liver during OTA + CTN + RSV treatment. However, OTA + CTN (0.125 + 20 and 0.250 + 20 mg kg−1) treatment resulted in a signifi- cant increase of CTN concentrations both in the kidney (9.71 ± 0.79, 13.67 ± 0.87 g g−1 tissue) and in the liver (7.02 ± 1.13, 8.33 ± 1.28) as compared to CTN-treated animals (2.23 ± 1.50, 1.76 ± 0.59). Resveratrol treatment counteracted the accumulation of CTN only in the kidney at a higher dose of OTA (0.250 mg kg−1) + CTN (20 mg kg−1). The OTA concentration in the brain correlated with the OTA dose, and it did not change after OTA + CTN treatment. The brain CTN concentration in OTA + CTN-treated animals showed the same pattern as in the liver and kidney.
