Sažetak
The cell is a morphological unit with a specific function in the body. It contains the nucleus and cytoplasm, as visualized by light microscopy. Cellular organelles can be visualized by electron microscopy. Immunohistochemistry is a method that enables antigen visualization through binding with a specific antibody and eliciting an immune response. The basis for any successful histologic technique is appropriate staining of optimally fixed tissue and a thin tissue section. Fixation is the first and most important procedure for appropriate preservation of cells and tissues. Properly fixed tissue maintains its structure and allows for the preparation of high quality histological slides. The aim of the fixation is to prevent degenerative changes in the tissue caused by ischemia, as well as to enhance structural stability of the processed tissue. Fixation prevents autolytic changes caused by cellular enzymes an also inhibits bacterial and mold overgrowth causing decay. Using practical knowledge and experience, a histotechnician (baccalaureus of medical laboratory diagnosis) plays a key role in the meticulous and delicate process of immunohistochemical staining by choosing the fastest and most suitable method to achieve the high quality final immunohistochemical slides.
