Sažetak
Background: Exposure of Campylobacter jejuni to sub-inhibitory concentrations of antibiotics due to inappropriate treatment can promote the bacterial resistance. Nevertheless the impact of such antimicrobial misuse on bacterial virulence potential is not yet understood. As the prevalence of antibiotic resistance is inclining, natural products with antimicrobial activity are becoming an attractive alternative for treatment of infections. One of such products is pinocembrin, present in honey and therefore a part of normal healthy diet in low concentrations. Methods: We evaluated the anti-Campylobacter activity of pinocembrin using broth microdilution and investigated how treatment of Campylobacter with sub-inhibitory doses (16 mg/L) of pinocembrin affects its virulence potential in systemic infection in BALB/c mice model. To better understand the bacterial response to treatment with sub-inhibitory concentrations of pinocembrin we have analysed the whole-genome transcriptomic response of C. jejuni to such treatment using the microarray approach. Results: Pinocembrin acted antimicrobially at the median concentration of 39 mg/L on 31 C. jejuni isolates. The 2-hour pre-treatment of C. jejuni with sub-inhibitory concentration of pinocembrin resulted in 1 log decrease in bacterial CFU counts of pinocembrin pre-treated culture compared to the untreated culture (p=0.024) in mice livers 8 days post- intravenous infection of mice. The pre-treatment evoked up-regulation of several NADH dehydrogenase I chain sub-units and proteins involved in iron uptake, suggesting the disruption of redox cycle and iron metabolism, which may cause the disadvantage in the host environment during the infection. Conclusions: Treatment of C. jejuni with pinocembrin inhibits its growth and decreases its virulence.
