Sažetak
In pharmaceutical drug analysis, the HPLC methods are used for analyzing raw materials using in drug synthesis as well as for identification of intermediates in drug synthesis, for off-line monitoring of reactions, for analyzing drug formulations, impurities and degradation products, and so on. Most analyses are done using reverse-phase HPLC with UV detection. Liquid chromatography coupled with mass spectrometry (LC-MS) has become the method-of-choice for analytical support in different stages of quality control. Due to high sensitivity and selectivity LC-MS and LC-MS-MS have become the predominant method in bioassays and pharmacokinetic and metabolic studies, as well as in the structure elucidation of drug impurities and degradation products. Within this lecture, the development of HPLC method for monitoring acid-catalyzed conversion of compound A to compound B, which is a key intemediate in the synthesis of one antiinflammatory drug, will be presented. Another example of application of LC in drug analysis which will be presented, is the development of UPLC method for determining degradation products of medetomidine present in Dorbene vet® solution.2
