Sažetak
Antimicrobial activities of four species from the genus Globularia L. (Globularia alypum L., G. cordifolia L., G. meridionalis (Podp.) O. Schwarz and G. punctata Lapeyr.) were compared for the first time by using the cylinder-plate diffusion method and a two-fold broth-microdilution method in Müller-Hinton broth using 96-well microtiter plates. Investigations were done on methanolic extracts obtained by Soxhlet extraction from aerial plant parts. The broth-microdilution method was followed by subcultivation of all dilutions to the surface of the Müller-Hinton agar and was also evaluated spectrophotometrically with 2, 3, 5- triphenyltetrazolium chloride (TTC) as indicator of bacterial growth to provide a double control in assessing the minimal inhibitory concentration (MIC) of all samples. The bacterial strains included in the original analysis were chosen according to those previously reported to be susceptible to extracts of the medicinal plant G. alypum, namely: B. cereus ATCC 11778, B. subtilis ATCC 6633, E. coli ATCC 10536, E. faecalis ATCC 29212, K. pneumoniae MFBF-10402, P. aeruginosa ATCC 27853 and S. aureus ATCC 6538 in order to evaluate the medicinal potential of the three remaining species, whose antimicrobial activity hasn't been investigated so far. Since the results of both diffusion and microdilution methods showed that all tested Globularia species possess substantial antistaphylococcal activity, the investigation of MICs was expanded on nine other S. aureus strains, including five clinical methicillin-resistant S. aureus (MRSA) strains. Additionally, extracts were tested for inhibitory activity against the S. aureus enterotoxins and biofilm production. No significant difference in the antistaphylococcal effect was noticed between different species, or between methicillin- sensitive and methicillin-resistant S. aureus. Apart from the inhibitory activity against S. aureus, all four species showed low inhibitory activity against B. cereus. Only G. alypum possessed activity against P. aeruginosa, while G. punctata was the only species which inhibited B. subtilis, E. faecalis and E. coli growth. No influence on staphylococcal enterotoxins and biofilm production was observed. The results of this study confirm the previously published results considering the antimicrobial activity of G. alypum and provide new insights into the antimicrobial potential of related Globularia species, especially that of G. punctata.
