Sažetak
Human erythrocytes were studied with respect to cell membrane organization and dynamics, as assessed by the spin probes methyl ester of doxyl palmitate, MeFASL(m, n). Electron paramagnetic resonance (EPR) line shapes were deconvoluted in order to determine the spectral components. Relative contribution of spectral components to the whole spectrum was interpreted in the sense of relative membrane population area (RMPA) by domains, characterized by different molecular ordering and dynamics in the lipid phase of cell membranes. At least three types of spectral components (domains), characterised by different order parameters, were resolved in spectra, indicating lateral heterogeneity of the membrane lipids with respect to the motional restriction imposed in the local membrane organization. As the spin label attached to the spin probe was moved deeper towards the membrane central region, the membrane organization changed. The change was obseved not only in the molecular order and dynamics of particular component, but also in the relative contribution of spectral components to the whole spectrum. The results were discussed and interpreted in the light of membrane lipid lateral and transversal organisation.
