Sažetak
Malondialdehyde (MDA) is product of lipid peroxidation and is commonly used as marker of oxidative stress. In order to test urinary MDA as relevant marker of oxidative stress the aim of this study was to check on MDA stability in the urine as well as on intra- and inter-individual changes in the level of urinary MDA. The stability of MDA in urine samples (n=5) at room temperature was checked for 24 h, and its stability at -20 °C in urine samples (n=7) for 10 days. The intra- and inter-individual changes in urinary MDA were monitored in urine samples (n=7) for 6 days and 24h-fluctuantions in MDA level were followed in two urine samples. In urine samples level of MDA was determined spectrophotometrically by validated method based on thiobarbituric acid (TBA) assay at 532 nm on Agilent 8453 UV- Vis (Agilent Technologies, CA, USA). Our results show that MDA in urine is stable at room temperature for 24h and when kept at -20 °C is stable for 10 days. The level of urinary MDA among persons vary from 0.98 µM to 7.51 µM and variation in urinary MDA for a person from day to day is observed. Also, the 24h-fluctuation of urinary MDA was noted and is probably affected by food consumption. Since MDA is stable product of lipid peroxidation and sampling of urine is more readily accepted by participants further studies on urinary MDA as marker of oxidative stress are needed.
