Sažetak
Cytochrome P450s 4A11, 2C8, and 2C9 share arachidonic acid as a substrate with 4A11 forming 20-hydroxyeicosatetraeonic acid (20-HETE) and 2C8 and 2C9 forming epoxidation products, epoxyeicosatrienoic acids (EETs). Literature suggests that 20-HETE and EETs may play opposing autacoid roles in the vascular system. One 4A11 isoform, F434S, is associated with a higher incidence of hypertension. Using a mass spectrometry-based label free approach, we measured protein levels of both 2C8 and 2C9 as well as homozygotes and heterozygotes of 4A11 isoforms. Initial results suggest that F434S may be subject to differential post-translational regulation.
