Sažetak
Lipases (EC 3.1.1.3) catalyze the hydrolysis of triacylglycerols to release free fatty acids and glycerol. One of possible ways for biodiesel production could be the application of lipases. In this work a) initial reaction rate of p-nitrophenyl palmitate (pNPP) hydrolysis catalyzed by lipase from Thermomyces lanuginosus in organic media with the addition of emulsifier was investigated ; and b) biodiesel sinthesis from sunflower oil with the use of lipase, in batch and fed batch reactors were performed. Three different organic solvents (methanol, acetonitrile and ethanol) and two different emulsifiers, Triton X-100 (50 v/v) and Tween 80 (1, 10, 20 and 30 w/v) were used in kinetic measurements. Three different lipase concentrations (2.5, 5 i 10 % w/v ; 0, 01 M phosphate buffer, pH 7.4) were investigated. Simple Michaelis-Menten kinetic model was developed and kinetic parameters (Km i Vm) were estimated by nonlinear regression using Scientist software. It was shown that Tween 80 can not be used for the measurement of kinetics of investigated reaction. Conversion up to 88% and 31% were reached after 24 hours for batch and fed-batch experiments, respectively.
