Sažetak
Dalmatian sage (Salvia officinalis L.) is native to the east side of Adriatic and Ionian seas. This perennial plant is widespread in Croatia, where it spontaneously grows on the sunny hillsides of the Dalmatian islands and adjacent coastal zones of the Adriatic Sea. In the northern part of the Adriatic coast, this species is very abundant, representing the only plant growing on the poor, rocky soils. This area is well known for its famous unifloral Salvia officinalis honey, widely used in traditional medicine. The objective of the present study was to find potential chemical markers for the authentication of unifloral Salvia officinalis honey. Research was mainly based on the analysis of the phenolic acids, their derivatives and flavonoids and on identification of target compounds using high resolution LC/MS techniques. Quantification of major phenolic compounds was achieved using ultra high-performance liquid chromatography coupled with diode array detector and triple quadruple mass spectrometer (UHPLC DAD-MS/MS). To trace phytochemicals that characterize sage honeys produced in Croatia UHPLC coupled with hybrid mass spectrometer which combines the Linear Trap Quadrupole and OrbiTrap mass analyzer (LTQ OrbiTrap MS) was utilized. A total of 25 compounds were quantified by comparing retention times and MS spectra with available standards. Based on the exact mass search and the study of the fragmentation pathways it was possible to individuate 36 compounds. Phenolic diterpenes, carnosol and carnosic acid, although present in sage, were not previously detected in sage unifloral honey. In this study, these compounds were identified in honey samples, but in trace amounts. Additionally, three types of available unifloral honeys of Lamiaceae species from the same region were introduced into the analysis: thyme (Thymus spp.) honey, mint (Mentha spp.) honey and winter savory (Satureja montana L.) honey to demonstrate the applicability of developed method for the authentication of sage unifloral honey. Principal Component Analysis (PCA) was employed to analyze semiquantitative data for 61 compounds in order to examine their relative variations within honeys of Lamiaceae species.
