Sažetak
New acidic method for determination of copper in blood serum by FAAS was elaborated. Flame AAS methods are still preferred for everyday clinical laboratory routine analyses of essential metals in biological samples. The optimum dilution of the sample analyzed by FAAS is the one that does not clog the capillary and burner head but still yields and adequate signal. For example, the reference method for Cu in blood serum is based on simple 2-fold dilution of serum with water (1), whereas Meret and Henkin (2) and Pizent and Telišman (3) used 6% n-butanol for enhancing the signal in 10-times diluted sera for 30%. The latter methods seem to be sufficiently reliable for analyzing sera with high or physiological level of Cu but their reliability for analyzing low-Cu sera may still be questionable. The method proposed in this report includes low dilution level (2-fold) and efficient deproteinization of serum with 1 mol L-1 HCl. This way we afforded the following major benefits of the method such as preserving the analytical signal and eliminating the risk of capillary clogging. For the sake of objective evaluation acidic systems pertaining to the new method amd aqueous systems pertaining to the reference method were statistically analyzed and compared within the working range of 0.05-2.0 microgram mL-1 Cu ; both calibration and standard additions have been taken into account. The new method has been demonstrated as sensitive, selective, precise (measurement repeatability being 1.8-5.1 and intermediate precision 8.5%) and highly accurate: the mean relative error and recovery values being +0.2% and 100.1%, resp. In respect to LOD (0.03 microgram mL-1), accuracy and precision the newly proposed acidic method has been found not significantly different from the reference one. Excellent agreement of the results for concentration of Cu in original serum of 1.22+-0.02 microgram mL-1 (mean+-SD, n = 5) was found by both methods. Moreover, no statistically significant differences for copper concentration were found within the acidic and aqueous groups of data no matter whether calibration or standard addition was used: mean recovery found was 100.8+-1.5% (n = 3. Standard additions were found redundant and might be reliably replaced by calibrations in 1 mol L-1 HCl enabling simple and rapid routine analyses of blood sera, particularly those with pathologically low copper.
